Sclerostin, secreted by osteocytes, is a key physiological inhibitor of osteogenesis. Pharmacologic inhibition of sclerostin using sclerostin‐neutralizing monoclonal antibody (Scl‐Ab) thus increases bone formation, bone mass and bone strength in models of osteopenia and fracture repair. Evenity is a bone-forming monoclonal antibody designed to inhibit the action of sclerostin, a regulatory factor in bone metabolism. This allows the drug to rapidly increase bone formation and, to a lesser extent, decrease bone resorption. Evenity is administered by subcutaneous injection once a month. Romosozumab (AMG 785) is the first humanized anti-sclerostin monoclonal antibody that has been demonstrated to increase bone formation.
